    Summary

    Nicotine alone significantly increased gastric ulcers in rats. However, combining nicotine with vitamin C (1-ascorbic acid) reduced ulcer incidence and severity, indicating no synergistic effect on stress-induced ulcers.

    Background:

    • Stress is a known factor in the development of gastric ulcers.
    • Nicotine is a common substance with potential gastrointestinal effects.
    • Vitamin C (1-ascorbic acid) is an antioxidant with potential protective properties.

    Purpose of the Study:

    • To investigate the individual and combined effects of nicotine and 1-ascorbic acid on stress-induced gastric ulcer formation in rats.
    • To determine if 1-ascorbic acid modifies nicotine-induced gastric ulceration.
    • To assess the synergistic potential of 1-ascorbic acid and nicotine in exacerbating stress-induced gastric ulcers.

    Main Methods:

    • Rats were orally administered 1-ascorbic acid, nicotine, a combination of both, or distilled water for 10 days.
    • Following treatment, rats were subjected to a 24-hour fast and a 2-hour restraint in a cold environment to induce stress.
    • Gastric ulcer incidence and severity were assessed.

    Main Results:

    • Nicotine administration alone significantly increased the number of gastric ulcers compared to controls.
    • 1-Ascorbic acid administration alone also increased ulceration relative to distilled water controls.
    • The co-administration of 1-ascorbic acid and nicotine resulted in a significant reduction in both the incidence and severity of gastric ulcers.

    Conclusions:

    • Nicotine alone exacerbates stress-induced gastric ulcers in rats.
    • 1-Ascorbic acid alone appears to increase ulcer susceptibility under these experimental conditions.
    • The combination of 1-ascorbic acid and nicotine does not act synergistically to worsen stress-induced gastric ulcers; instead, it shows a protective effect against ulceration.
