    Summary

    Ethanol reduces complex spike activity in cerebellar Purkinje cells and antagonizes harmaline-induced tremor. Its effects on Purkinje cells suggest a mechanism beyond the climbing fiber synapse, involving the inferior olive.

    Background:

    • Ethanol is known to affect cerebellar function.

    Purpose of the Study:

    • To investigate the effects of ethanol on cerebellar Purkinje cell activity.
    • To determine the mechanism by which ethanol antagonizes harmaline-induced tremor.

    Main Methods:

    • Intravenous administration of ethanol and harmaline in rats.
    • Electrophysiological recording of Purkinje cell complex spike (CS) activity.
    • Juxtafastigial stimulation to evoke CS.

    Main Results:

    • Ethanol (1.5 g/kg i.v.) decreased spontaneous CS activity in Purkinje cells without altering the threshold for evoked CS.

    • Ethanol's reduction of spontaneous CS activity is likely due to its depressant effects on the inferior olive, not a direct action on the climbing fiber-Purkinje cell synapse.
    • The inferior olive's depression by ethanol contributes to, but does not fully explain, the blockade of harmaline tremor.
