    Background:

    • Previous studies noted ergotamine decreases peripheral-central systolic blood pressure (SBP) gradients.
    • The mechanism behind this SBP decrease, whether intra-arterial or due to vessel wall tone, remained unclear.

    Purpose of the Study:

    • To investigate the effect of ergotamine tartrate on finger-arm systolic gradients.
    • To determine if ergotamine acts peripherally to reduce SBP.

    Main Methods:

    • Administered ergotamine tartrate intravenously (0.5 mg) and intra-arterially (10 µg).
    • Measured finger-arm systolic gradients following arterial infiltration with papaverine or dihydralazine.
    • Assessed gradients with cuff occlusion of finger blood supply before and after ergotamine administration.

    Main Results:

    • A normal reduction in finger-arm systolic gradients was observed.
    • Intravenous and intra-arterial ergotamine tartrate produced similar decreases in gradients.
    • The findings suggest a peripheral site of action for ergotamine's effect on SBP.

    Conclusions:

    • Ergotamine tartrate induces a decrease in peripheral intra-arterial SBP.
    • The primary mechanism is likely the contraction of arteries proximal to the digital arteries.
    • Ergotamine's hypotensive effect is mediated by peripheral arterial constriction.
