    Background:

    Purpose of the Study:

    • To investigate platelet serotonin uptake characteristics in migraine patients.

    Main Methods:

    • Platelet-rich plasma was obtained from 11 migraineurs and 15 healthy volunteers.
    • Platelet serotonin uptake kinetics (Km, Vmax, y) were measured.
    • Migrainous platelets were tested in isolation and after resuspension in control plasma.

    • Platelet serotonin uptake parameters (Km, Vmax, y) were significantly decreased in migraineurs experiencing an active migraine attack during blood collection.
    • Platelet serotonin uptake normalized when migrainous platelets were isolated or resuspended in normal plasma.
    • No significant differences in uptake were observed between migraineurs without an active attack and healthy controls.

    Conclusions:

    • The decrease in platelet serotonin uptake during migraine attacks is likely mediated by plasmatic factors, not an intrinsic platelet abnormality.
    • This finding suggests that circulating factors during a migraine attack reversibly impair platelet serotonin transport.
    • Further research into these plasmatic factors may reveal novel diagnostic or therapeutic targets for migraine.
