Acute solvent-ethanol interactions with special reference to xylene

V Riihimäki, A Laine, K Savolainen

Scandinavian Journal of Work, Environment & Health
    |March 1, 1982

    Summary

Ethanol ingestion significantly impairs the body's ability to process m-xylene, a common industrial solvent. This interaction can lead to inaccurate biological monitoring of solvent exposure and cause adverse reactions in sensitive individuals.

    Area of Science:

    • Occupational Health
    • Toxicology
    • Environmental Health

    Background:

    • Ethanol is known to inhibit the metabolism of industrial solvents like trichloroethylene and dimethylformamide.
    • Solvent exposure can interfere with ethanol metabolism, leading to acetaldehyde accumulation and alcohol intolerance symptoms.

    Purpose of the Study:

    • To investigate the metabolic interaction between acute ethanol ingestion and subsequent m-xylene inhalation exposure.
    • To determine the impact of ethanol on m-xylene clearance and biological monitoring accuracy.

    Main Methods:

    • Subjects received a single dose of ethanol (0.8 g/kg).
    • Following ethanol ingestion, subjects were exposed to m-xylene via inhalation (6.0 & 11.5 mmol/m3 for 4 hours).
    • Metabolic clearance, blood concentrations, and urinary excretion of methylhippuric acid were measured.

    Related Experiment Videos

    Main Results:

    • Ethanol significantly impaired the metabolic clearance of m-xylene.
    • Blood m-xylene concentrations were elevated following combined exposure.
    • Urinary excretion of methylhippuric acid, a biomarker for m-xylene uptake, was decreased.
    • Some individuals experienced nausea and dermal flush from combined exposure.

    Conclusions:

    • Ethanol ingestion is a significant confounding factor in the biological monitoring of m-xylene exposure.
    • The combined exposure of ethanol and m-xylene can lead to adverse health effects in susceptible individuals.
    • Occupational health guidelines should consider the impact of ethanol consumption on solvent exposure assessment.
