    Summary

    Long-term goitrogen administration significantly increases thyroid weight and stimulates thyroid-stimulating hormone (TSH) levels. This study reveals mast cell hyperplasia occurs during goitre formation in rats.

    Background:

    • Goitre is characterized by thyroid gland enlargement.
    • The role of mast cells in thyroid physiology is not fully understood.
    • Goitrogens disrupt thyroid hormone synthesis, leading to adaptive changes.

    Purpose of the Study:

    • To investigate the impact of long-term goitrogen administration on rat thyroid mast cell populations.
    • To quantitatively assess changes in mast cell number, size, and distribution during goitre formation.
    • To explore the relationship between thyroid-stimulating hormone (TSH) levels and mast cell proliferation.

    Main Methods:

    • Rats were administered the goitrogen aminotriazole to block thyroid hormone synthesis.
    • Thyroid tissue was analyzed after 80 days of treatment and compared to controls.
    • Serum TSH levels were measured using radioimmunoassay.
    • Mast cell populations were quantified via histological staining and microscopic analysis.

    Main Results:

    • Goitrogen treatment resulted in a 9-fold increase in serum TSH and an 8-fold increase in thyroid weight.
    • While mast cell density per unit volume decreased, total mast cell numbers per gland increased 4-5 fold.
