Efficacy of a bacterin prepared from Chlamydia psittaci grown in cell culture for experimental immunization of ewes
Veterinary Microbiology
|November 1, 1982
Abstract:
The efficacy of a bacterin prepared from Chlamydia psittaci grown in mouse L-cells was compared to a similar bacterin prepared from C. psittaci grown in chicken embryos (CE). Both bacterins significantly reduced (P less than 0.03) incidence of abortion and weak lambs compared to non-vaccinated control ewes. The L-cell bacterin elicited a greater antibody response than the CE bacterin (P less than 0.03).
Insights
A new bacterin from L-cell grown Chlamydia psittaci (C. psittaci) and one from chicken embryos (CE) both reduced lamb abortions. The L-cell bacterin showed a superior antibody response in ewes.
Area of Science:
- Veterinary immunology
- Microbial pathogenesis
- Ovine reproductive health
Background:
- Chlamydia psittaci (C. psittaci) causes significant economic losses in sheep farming due to abortions and weak lambs.
- Current vaccines for C. psittaci often utilize chicken embryo (CE) derived bacterins.
- Alternative cell culture methods for antigen production may offer improved efficacy or immunogenicity.
Purpose of the Study:
- To compare the efficacy and immunogenicity of a C. psittaci bacterin derived from mouse L-cells against a CE-derived bacterin.
- To evaluate the impact of different bacterin production methods on ovine reproductive outcomes.
- To assess the antibody response elicited by L-cell versus CE bacterins.
Main Methods:
- Preparation of two bacterins: one from C. psittaci grown in mouse L-cells and another from C. psittaci grown in chicken embryos (CE).
- Vaccination of ewes with either the L-cell or CE bacterin, with a control group receiving no vaccination.
- Monitoring and comparison of abortion rates, incidence of weak lambs, and serum antibody titers between vaccinated and control groups.
Main Results:
- Both L-cell and CE bacterins significantly reduced the incidence of abortion and weak lambs compared to unvaccinated controls (P < 0.03).
- The L-cell bacterin elicited a significantly greater antibody response in ewes than the CE bacterin (P < 0.03).
- No significant difference in efficacy regarding abortion/weak lamb incidence was noted between the two bacterin types, but immunogenicity differed.
Conclusions:
- Bacterins prepared from C. psittaci grown in mouse L-cells are effective in reducing ovine abortions and weak lambs.
- L-cell derived C. psittaci bacterins induce a stronger antibody response compared to CE-derived bacterins.
- Mouse L-cell culture represents a viable alternative for producing effective C. psittaci vaccines for sheep, potentially offering enhanced immunogenicity.
