Multiple genes coding for octopine-degrading enzymes in Agrobacterium

A L Montoya, L W Moore, M P Gordon

    Journal of Bacteriology
    |December 1, 1978
    PubMed
    Summary

Most Agrobacterium strains that use nopaline also break down octopine. However, octopine degradation genes may be chromosomal, and virulence plasmids appear incompatible, suggesting a shared origin.

    Area of Science:

    • Microbiology
    • Plant Pathology
    • Molecular Biology

    Background:

    • Biotype 2 Agrobacterium tumefaciens and A. radiobacter strains often utilize nopaline and degrade octopine.
    • Understanding the genetic basis and transfer of these metabolic capabilities is crucial for plant disease management.

    Purpose of the Study:

    • To investigate the genetic location and transferability of octopine degradation genes in Agrobacterium.
    • To determine the regulation and compatibility of plasmids involved in octopine and nopaline utilization.

    Main Methods:

    • Plasmid transfer experiments to assess gene mobility.
    • Isolation of mutants to identify chromosomal vs. plasmid-borne genes.
    • Inducibility and constitutive expression studies of metabolic pathways.
    • Plasmid transformation assays to evaluate compatibility.

    Main Results:

    • Octopine degradation ability was not efficiently transferred with nopaline utilization genes.
    • Octopine degradation genes were found to have a chromosomal location in some strains.
    • Octopine degradation was inducible when plasmid-borne, but constitutive in strains degrading both substrates.
    • Plasmids conferring octopine utilization were incompatible with nopaline or null-type plasmids.

    Conclusions:

    • The incompatibility of virulence plasmids suggests a common evolutionary origin for major plasmid groups in Agrobacterium.
    • Genetic location and regulation of octopine degradation vary, with potential chromosomal involvement.
    • Plasmid incompatibility impacts the co-existence and transfer of metabolic traits.
