    Summary

    This study identifies four major urine ribonuclease (RNase) activities, with Band A being the most abundant and similar to serum RNase 1. Kidney processing differs for RNase 2-5 compared to RNase 1.

    Background:

    • Urine contains multiple ribonuclease (RNase) activities.
    • Previous studies have reported various RNase preparations in urine with differing characteristics.

    Purpose of the Study:

    • To characterize and compare the major urine RNase activities with known serum RNases.
    • To investigate the processing of RNases by the kidney.

    Main Methods:

    • Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with activity staining.
    • Gel filtration chromatography.
    • Phosphocellulose and Sephadex chromatography.

    Main Results:

    • Four major urine RNase bands (A-D) were identified with distinct pH optima and molecular weights.
    • Band A, the most abundant, resembles serum RNase 1.
    • Bands B and C are similar to serum RNase components, and Band D resembles leukocyte RNase-like activity.
    • Urine RNase 1 is more concentrated than serum RNase 1.
    • Urine counterparts of serum RNases 2, 4, and 5 are not readily detected, suggesting different kidney processing.

    Conclusions:

    • Urine RNase composition differs from serum, with Band A being a prominent RNase 1-like activity.
    • The kidney appears to process serum RNases 2-5 differently than RNase 1.
    • Existing literature on urine RNases can be reconciled with Bands A and D, but not with bovine pancreatic RNase A.
