    Background:

    • Androgens play crucial roles in male development and physiology.
    • Understanding tissue-specific androgen metabolism is vital for clinical applications.
    • Age-related changes in androgen levels are well-documented in circulation but less so in specific tissues.

    Purpose of the Study:

    • To quantify testosterone, DHT, and Adiol concentrations in various postmortem male skin and muscle tissues.
    • To assess the activity of 5 alpha-reductase and 3-reductase enzymes across these tissues.
    • To investigate potential age-dependent variations in tissue androgen levels.

    Main Methods:

    • Postmortem tissue samples (pubic skin, scrotal skin, thigh skin, striated muscle) were collected from 24 males (aged 20-82).
    • Concentrations of testosterone, 5 alpha-androstane-17 beta-ol-3-one (DHT), and 5 alpha-androstane-3 alpha,17 beta-diol (Adiol) were measured.
    • Enzyme activities were inferred using ratios of specific androgen metabolites.

    Main Results:

    • Androgen concentrations were highest in scrotal skin, followed by pubic skin, and lowest in thigh skin and striated muscle.
    • 5 alpha-reductase activity was highest in scrotal skin, decreasing in pubic, thigh, and muscle tissues.

    • Scrotal skin maintains high androgen levels independent of age, potentially due to embryological similarities with the prostate.
    • Pubic skin exhibits age-related androgen decline, unlike scrotal skin, thigh skin, or striated muscle.
