    Background:

    • Atherosclerosis involves changes in the extracellular matrix of arterial walls.
    • Collagen composition is altered in atherosclerotic plaques.

    Purpose of the Study:

    • To investigate the relative abundance of alpha chains in collagen within human atherosclerotic plaques.
    • To compare collagen composition between atherosclerotic plaques and normal aortic tissue.

    Main Methods:

    • Pepsin-solubilized collagens were extracted from 28 human aortas.
    • Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was used to estimate the relative abundance of alpha chains.
    • The ratio of alpha B chain (Type V collagen) to alpha 1(I) chain was quantified.

    Main Results:

    • The ratio of alpha B chain to alpha 1(I) chain was significantly increased in atherosclerotic plaques compared to non-sclerotic aorta.
    • This indicates a relative increase in Type V collagen or its alpha B chain component in plaque tissue.

    Conclusions:

    • Proliferating smooth muscle cells in atherosclerotic lesions may synthesize more alpha B chains of Type V collagen.
    • Altered collagen synthesis by smooth muscle cells is implicated in human atherogenesis.
    • These findings contribute to understanding the molecular changes in atherosclerotic plaque development.
