    Summary

    Exogenous progesterone did not alter post-term fetal lung growth in rats. Prolonged gestation, whether induced by progesterone or surgical retention, showed similar adverse effects on fetal development and lung maturation.

    Background:

    • Previous studies demonstrated progesterone's ability to prolong gestation in rats.
    • Understanding the impact of prolonged gestation on fetal lung development is crucial.

    Purpose of the Study:

    • To investigate the effects of exogenous progesterone on post-term fetal lung growth.
    • To compare the effects of progesterone-induced prolonged gestation with surgically retained post-term fetuses on lung development.

    Main Methods:

    • Surgically retained fetuses for 1-3 days beyond term.
    • Harvested fetuses via caesarean section between gestation days 21-25.
    • Analyzed fetal body, organ weights, lung histology, glycogen, DNA, and disaturated phosphatidylcholine (DSPC).

    Main Results:

    • Both retained and P20 post-term fetuses showed increased body, kidney, spleen weights, reduced lung/liver weights, lung atelectasis, and hypoglycemia.
    • Retained fetuses did not exhibit changes in lung DSPC and DNA content post-term, unlike P20 fetuses.
    • P20 fetuses displayed larger adrenals with prolonged gestation.

    Conclusions:

    • Exogenous progesterone does not appear to influence fetal lung growth post-term.
    • Fetal distress, rather than progesterone, may negatively impact lung cellularity and DSPC content in post-term fetuses.
