    Summary

Guinea pig thymic fragments in organ culture lost lymphocytes and their epithelium transformed into keratinized squamous epithelium. This suggests Hassall

    Area of Science:

    • Histology and Cell Biology
    • Immunology
    • Developmental Biology

    Background:

    • The thymus is a primary lymphoid organ crucial for T-cell maturation.
    • Thymic epithelial cells play a vital role in supporting lymphocyte development.
    • Hassall's corpuscles are characteristic structures within the thymic medulla, but their origin and function are not fully understood.

    Purpose of the Study:

    • To investigate the morphological changes of guinea pig thymic fragments during in vitro organ culture.
    • To elucidate the transformation of thymic epithelium and the origin of Hassall's corpuscles.
    • To analyze cell proliferation and differentiation within the cultured thymic tissue.

    Main Methods:

    • In vitro organ culture of guinea pig thymic fragments.
    • Sequential morphological analysis using light and electron microscopy.
    • 3H-thymidine autoradiography to track cell proliferation and distribution.

    Main Results:

    • Cultured thymic fragments progressively lost lymphocytes, becoming alymphoid.
    • Thymic epithelium transformed into keratinized squamous epithelium, originating from corticomedullary areas.
    • Keratinization initiated around Hassall's corpuscles, leading to their enlargement and eventual complete keratinization of fragments.

    Conclusions:

    • Hassall's corpuscles are derived from keratinized thymic epithelium.
    • The formation and existence of Hassall's corpuscles are linked to the normal function of thymic epithelial cells.
    • Organ culture provides a model to study thymic epithelial cell differentiation and keratinization.
