    Area of Science:

    • Cardiovascular Physiology
    • Molecular Cardiology
    • Ion Channel Biophysics

    Background:

    • The rapid inward sodium current (INa) is critical for cardiac action potential generation.
    • Understanding INa kinetics is essential for comprehending normal heart function and disease states.

    Purpose of the Study:

    • To comprehensively characterize the biophysical properties of INa in isolated rat ventricular myocytes.
    • To elucidate the activation, inactivation, and recovery kinetics of the cardiac sodium channel.

    Main Methods:

    • Utilized the double-suction-pipette voltage clamp technique on isolated rat ventricular cells.
    • Investigated INa using voltage steps from a holding potential of -80 mV at 20-22°C.
    • Analyzed current-voltage relationships, conductance, activation, inactivation, and recovery time constants.

    Main Results:

    • INa exhibited a threshold between -70 and -60 mV, peaking at -30 to -20 mV.
    • Activation time constants ranged from 0.7 ms to 100 µs, while inactivation required two time constants (τh1, τh2).
    • Recovery from inactivation also showed two time constants (τr1, τr2), with the slower component around 100 ms.

    Conclusions:

    • The study provides detailed kinetic parameters for rat ventricular INa, including activation and inactivation dynamics.
    • Cardiac INa demonstrated low sensitivity to tetrodotoxin, requiring 30 µM for complete blockade.
    • These findings contribute to a deeper understanding of cardiac sodium channel function and its role in electrophysiology.
