    Summary

    Epicutaneous skin testing with 1-monochlorine-2,4-dinitrobenzene (DNCB) effectively induces cell-conditioned immunity, provable by late-type allergic skin reactions. Self-produced DNCB skin test plasters are suitable for clinical use.

    Background:

    • Cell-conditioned immunity can be induced via epicutaneous skin testing.
    • 1-monochlorine-2,4-dinitrobenzene (DNCB) is a chemical agent used for inducing such immune responses.

    Purpose of the Study:

    • To confirm the clinical suitability of self-produced DNCB skin test plasters.
    • To evaluate the efficacy of DNCB in inducing and detecting cell-conditioned immunity.

    Main Methods:

    • Epicutaneous skin testing using DNCB-layered foil plasters on 125 healthy individuals (age 17-57).
    • Application of 0.049 molar DNCB solution (1,000 µg DNCB: 100 µl olive oil) with a 48-hour skin contact period.
    • Assessment of late-type allergic skin reactions and eczema-typical symptoms for characterization and quantification.

    Main Results:

    • A 48-hour DNCB skin contact induced late reactions in 84% of cases, increasing to 98% upon repeat testing.
    • Eczema-typical symptoms proved suitable for characterizing and quantifying immune responses.
    • No significant differences were observed based on age or sex; ultrastrong reactions were not recorded.

    Conclusions:

    • Self-produced DNCB skin test plasters are clinically suitable for inducing and assessing cell-conditioned immunity.
    • The DNCB epicutaneous test is an effective method for evaluating delayed-type hypersensitivity.
    • The method allows for reliable characterization and quantification of immune responses without significant adverse reactions.
