    Summary

Experimental immunologic tolerance induced by BCG and cyclophosphane in mice prevented the development of tuberculosis immunity. Tolerant mice failed to mount cellular immune responses and lacked protection against virulent Mycobacterium tuberculosis infection.

    Area of Science:

    • Immunology
    • Microbiology
    • Infectious Diseases

    Background:

    • Tuberculosis remains a significant global health challenge.
    • Bacillus Calmette-Guérin (BCG) vaccination is the primary vaccine against tuberculosis.
    • Understanding the mechanisms of BCG-induced immunity is crucial for vaccine development.

    Purpose of the Study:

    • To investigate the impact of experimental immunologic tolerance on BCG-induced immunity and resistance to tuberculosis.
    • To determine if BCG vaccination confers protection in a state of suppressed immune responsiveness.

    Main Methods:

    • Induction of immunologic tolerance in CBA mice using a high dose of BCG and cyclophosphamide.
    • Assessment of cellular immunity through blast transformation and skin tuberculin tests.

    Related Experiment Videos

  • Challenge with virulent Mycobacterium tuberculosis H(37)Rv to evaluate resistance.
  • Main Results:

    • Tolerant mice did not develop cellular immunity to tuberculosis post-BCG vaccination.
    • Blast transformation and skin tuberculin tests indicated a lack of immune response in tolerant animals.
    • BCG vaccination failed to provide a protective effect against virulent M. tuberculosis infection in tolerant mice.

    Conclusions:

    • Experimental immunologic tolerance abrogates BCG-induced cellular immunity against tuberculosis.
    • Suppressed immune responses prevent the protective efficacy of BCG vaccination.
    • These findings highlight the critical role of a robust immune response for effective tuberculosis vaccination.
