Probucol in hypercholesterolemia. A double blind study

W F Riesen, M Keller, R Mordasini

    Atherosclerosis
    |June 1, 1980
    PubMed
    Summary

Probucol effectively lowers total and LDL-cholesterol in patients with primary hypercholesterolemia, alongside a decrease in apolipoprotein B. However, it also significantly reduces HDL-associated apolipoproteins A1 and A2.

    Area of Science:

    • Cardiology
    • Pharmacology
    • Biochemistry

    Background:

    • Primary hypercholesterolemia is a significant risk factor for cardiovascular disease.
    • Lipid-lowering therapies aim to reduce cardiovascular events.
    • Understanding the effects of specific drugs on lipoprotein profiles is crucial for effective treatment.

    Purpose of the Study:

    • To evaluate the efficacy of Probucol in managing serum lipids and lipoproteins.
    • To assess the impact of Probucol on apolipoproteins A1, A2, and B in hypercholesterolemic patients.

    Main Methods:

    • A double-blind, placebo-controlled study involving 27 patients with primary hypercholesterolemia.
    • Patients received Probucol (1000 mg/day) or placebo for 4 months after a 3-month diet and 6-week placebo period.
    • Measurements included serum lipids, lipoproteins, and apolipoproteins A1, A2, and B.

    Main Results:

    • Probucol significantly reduced total cholesterol (13%) and LDL-cholesterol (16%).
    • Apolipoprotein B decreased by 12%.
    • HDL cholesterol, triglycerides, and triglyceride-rich lipoproteins were not significantly altered, but apolipoproteins A1 and A2 were markedly reduced.

    Conclusions:

    • Probucol demonstrates efficacy in reducing LDL-cholesterol and apolipoprotein B in hypercholesterolemic patients.
    • The drug's significant reduction of HDL-associated apolipoproteins A1 and A2 warrants further investigation regarding its overall cardiovascular benefit.
    • Probucol was generally well-tolerated by patients.
