    Background:

    • Spiroplasma are helical, wall-less bacteria with unique genetic elements.
    • The presence and nature of extrachromosomal DNA in Spiroplasma have not been extensively characterized.

    Purpose of the Study:

    • To investigate the occurrence and characteristics of circular deoxyribonucleic acid (DNA) molecules in Spiroplasma strains.
    • To determine if these circular DNA molecules are viral or plasmid in nature.

    Main Methods:

    • Ethidium bromide-cesium chloride gradient centrifugation of Spiroplasma cell lysates.
    • Electron microscopy and agarose gel electrophoresis for analyzing DNA molecules.
    • Quantification of circular DNA relative to total cellular DNA.

    Main Results:

    • Ten out of twelve Spiroplasma strains contained multiple covalently closed circular DNA molecules.
    • Detected circular DNA molecules ranged in size from 1 x 10^6 to 26 x 10^6 Daltons, with 2-8 size classes per strain.
    • Common size classes were observed across different strains, and circular DNA constituted up to 12% of total cellular DNA.

    Conclusions:

    • The circular DNA molecules found in Spiroplasma are likely plasmids, not viral genomes or replicative forms.
    • The presence of these potential plasmids appears independent of Spiroplasma virus carriage or production.
