    Background:

    • Histamine is a key mediator of allergic skin reactions, causing itch and erythema.
    • Prostaglandin E2 (PGE2) is also involved in inflammatory processes, but its interaction with histamine in skin responses is not fully understood.

    Purpose of the Study:

    • To investigate the individual and combined effects of histamine and PGE2 on itch and erythematous responses in human skin.
    • To determine if PGE2-induced responses are mediated by histamine release.
    • To assess the potentiation effect of PGE2 on histamine-induced reactions.

    Main Methods:

    • Intradermal injections of histamine and PGE2 were administered to human subjects.
    • Subjects were pretreated with the antihistaminic drug chlorcyclizine.
    • Itch and flare responses were quantified and compared under different conditions.

    Main Results:

    • Both histamine and PGE2 induced itch and flare responses.
    • Chlorcyclizine pretreatment alleviated responses to both histamine and PGE2, suggesting histamine release involvement in PGE2 effects.
    • Combined histamine and PGE2 elicited prolonged itch and larger flare areas than additive effects, indicating potentiation.

    • PGE2 contributes to itch and erythema in human skin, at least partly via histamine release.
