    Summary

Whole-body irradiation in rats significantly reduced the clearance of gentamicin, an antibiotic, by the seventh day post-exposure. Concurrently, lung absorption of this drug increased following radiation.

    Area of Science:

    • Pharmacokinetics and Radiation Biology

    Background:

    • Gentamicin is a critical antibiotic for treating bacterial infections.
    • Radiation exposure can alter drug metabolism and elimination.
    • Understanding radiation effects on antibiotic pharmacokinetics is vital for patient care.

    Purpose of the Study:

    • To investigate the impact of whole-body irradiation on gentamicin clearance in rats.
    • To assess changes in gentamicin absorption from the lungs post-irradiation.

    Main Methods:

    • Rats received a single whole-body irradiation dose (6 Gy 60Co).
    • Intravenous and intratracheal administration of gentamicin was performed.
    • Drug clearance rates and lung absorption were monitored post-irradiation.

    Main Results:

    • A significant decrease in gentamicin clearance was observed after irradiation.
    • The lowest clearance rate occurred on the 7th day post-irradiation.
    • An increased absorption rate of gentamicin from the lungs was noted.

    Conclusions:

    • Single whole-body irradiation impairs gentamicin elimination in rats.
    • Radiation exposure enhances gentamicin absorption from pulmonary tissues.
    • These findings highlight potential therapeutic challenges in irradiated patients.
