    Summary

This study found that endocrine abnormalities, including adrenal and thyroid insufficiency, affect multiple siblings and a mother. Early screening can identify relatives at risk for developing these serious hormone deficiencies.

    Area of Science:

    • Endocrinology
    • Genetics
    • Autoimmune diseases

    Background:

    • Investigated a kindred with a high incidence of endocrine disorders.
    • Focused on adrenal and thyroid insufficiency within a family.
    • Explored the genetic and autoimmune basis of endocrine dysfunction.

    Observation:

    • Overt adrenal and thyroid insufficiency observed in three brothers from 17 offspring.
    • Endocrine abnormalities detected in 8 of 12 examined siblings and one mother.
    • Associated conditions included pernicious anemia, vitiligo, and alopecia.

    Findings:

    • Early compensated adrenal and thyroid hypofunction identified in asymptomatic relatives.
    • Presence of organ-specific antibodies in individuals with subclinical disease.
    • Clinical onset of endocrine deficiencies ranged from 18 to 38 years.

    Implications:

    • Highlights the importance of familial investigations for endocrine disorders.
    • Suggests genetic predisposition to autoimmune endocrine diseases.
    • Early identification of at-risk asymptomatic individuals can prevent severe deficiencies.
