    Summary

The heritable-translocation test in mice requires improvement for practical use. While 10 chemicals, primarily alkylating agents, induce translocations, standardization and cost-effective controls are crucial for reliable genetic testing.

    Area of Science:

    • Genetics
    • Toxicology
    • Mammalian Genetics

    Background:

    • The heritable-translocation test in mice is a key assay for detecting genetic damage.
    • Its utility in practical risk assessment is under continuous evaluation.
    • Evaluating existing literature is essential for understanding assay performance.

    Purpose of the Study:

    • To assess the practical utility of the heritable-translocation test in mice.
    • To review and evaluate data from published studies on chemical-induced heritable translocations.
    • To identify areas for improvement in the test's methodology and application.

    Main Methods:

    • Literature review of 47 reports on the heritable-translocation test in mice.
    • Evaluation of data adequacy for 32 compounds.

    Related Experiment Videos

  • Classification of chemical effects as positive, negative, or suggestive.
  • Main Results:

    • Data were adequate for 29 of 47 reports, covering 32 compounds.
    • 10 chemicals were confirmed to induce heritable translocations, mainly alkylating agents.
    • 3 compounds showed suggestive evidence, and data for 15 were inadequate.

    Conclusions:

    • The heritable-translocation test needs methodological standardization and validation.
    • The use of historical controls requires further investigation due to the prohibitive cost of concurrent controls.
    • Research into non-alkylating clastogens and error size in translocation heterozygote classification is needed.
