Basic fibroblast growth factor (bFGF) increases tenascin expression in astrocytes, potentially hindering central nervous system regeneration after injury. This effect is transient, with tenascin levels returning to normal after bFGF removal.
Area of Science:
- Neuroscience
- Cell Biology
- Biochemistry
Background:
- Extracellular matrix (ECM) molecules regulate neuronal development and injury response.
- Astrocytes exhibit heterogeneous tenascin expression, impacting neurite outgrowth.
- Basic fibroblast growth factor (bFGF) can reduce neuronal growth and adhesion on astrocytes.
Purpose of the Study:
- To investigate if bFGF increases tenascin expression in rat cerebral cortical astrocytes.
- To understand the role of bFGF-induced tenascin in central nervous system (CNS) regeneration.
Main Methods:
- Primary rat cerebral cortical astrocytes were cultured and treated with varying concentrations of bFGF.
- Tenascin levels were quantified using Western blot analysis and immunocytochemistry.
- Effects of serum, heparin, and treatment duration on tenascin expression were assessed.
Main Results:
- bFGF treatment significantly increased tenascin levels in a dose-dependent manner.
- Tenascin expression increased within 24-48 hours and persisted for 8 days.
- Serum and heparin enhanced bFGF's effect on tenascin production, which was reversible upon bFGF withdrawal.
Conclusions:
- bFGF induces tenascin expression in astrocytes, potentially creating an environment unfavorable for CNS regeneration.
- The transient nature of bFGF's effect suggests a dynamic regulation of the glial scar.
- Understanding this mechanism may offer therapeutic targets for promoting CNS repair.
