Summary

Lung fibroblasts from fibrotic lung tissue express acidic fibroblast growth factor (aFGF). This growth factor stimulates fibroblast proliferation, suggesting a role for aFGF in the development of lung fibrosis.

Area of Science:

  • Cell Biology
  • Molecular Biology
  • Pulmonary Medicine

Background:

  • Lung fibrosis is a debilitating condition characterized by progressive scarring of lung tissue.
  • Fibroblast activation is a key process in the pathogenesis of lung fibrosis.
  • The role of specific growth factors in fibroblast activation in lung fibrosis requires further elucidation.

Purpose of the Study:

  • To investigate the expression of growth factors in lung fibroblasts from a patient with lung fibrosis.
  • To determine the effect of acidic fibroblast growth factor (aFGF) on lung fibroblast proliferation.

Main Methods:

  • Isolation of a lung fibroblast cell line from a patient with lung fibrosis.
  • Reverse transcription-polymerase chain reaction (RT-PCR) to assess gene expression of ten growth factors.
  • Treatment of fibroblasts with recombinant aFGF and assessment of cell proliferation.

Main Results:

  • Lung fibroblasts expressed acidic fibroblast growth factor (aFGF) mRNA.
  • Recombinant aFGF was internalized by the fibroblasts.
  • aFGF stimulation led to increased fibroblast proliferation.

Conclusions:

  • Lung fibroblasts in fibrotic tissue express aFGF.
  • aFGF may activate lung fibroblasts in an autocrine manner.
  • aFGF is implicated as a potential mediator in the development of lung fibrosis.
