Summary

Galanin message-associated polypeptide (GMAP) does not inhibit insulin secretion from rodent islets, unlike its precursor galanin. This finding clarifies GMAP

Area of Science:

  • Endocrinology and Metabolism
  • Molecular and Cellular Biology

Background:

  • Galanin message-associated polypeptide (GMAP) is derived from preprogalanin.
  • GMAP-like immunoreactivity has been observed in pancreatic insulin cells.

Purpose of the Study:

  • To investigate if synthetic rat GMAP inhibits glucose-stimulated insulin secretion (GSIS) from rodent islets.
  • To compare the effects of GMAP and galanin on GSIS.

Main Methods:

  • Incubation of isolated rat and mouse islets.
  • Stimulation of insulin secretion with glucose (8.3 or 11.1 mM).
  • Treatment with varying concentrations of synthetic rat GMAP (0.1 nM to 1 microM) or rat galanin (100 nM).

Main Results:

  • GMAP did not alter glucose-stimulated insulin secretion across a broad dose range.

Related Experiment Videos

  • Rat galanin, at 100 nM, completely inhibited glucose-stimulated insulin secretion.
  • GMAP exhibits a differential effect on insulin secretion compared to galanin.
  • Conclusions:

    • GMAP does not appear to play a role in regulating insulin secretion from pancreatic islets.
    • The inhibitory effect of galanin on insulin secretion is specific and not replicated by GMAP.
