Summary

Flow cytometry revealed aneuploid DNA profiles in 29% of interstitial cystitis patients, suggesting potential epithelial cell abnormalities. DNA tetraploidy observed was attributed to binucleated cells, not true aneuploidy.

Background:

  • Interstitial cystitis (IC) is a chronic condition characterized by bladder pain and urinary urgency.
  • The cellular and molecular underpinnings of IC remain incompletely understood.
  • Investigating epithelial cell DNA content may offer insights into IC pathogenesis.

Purpose of the Study:

  • To analyze deoxyribonucleic acid (DNA) profiles of bladder epithelial cells from IC patients and controls using flow cytometry.
  • To identify potential DNA abnormalities, including aneuploidy and tetraploidy, in IC patient samples.
  • To explore the significance of observed DNA profiles in relation to IC.

Main Methods:

  • Flow cytometry was utilized to analyze bladder cell DNA content from 14 IC patients and 11 control subjects.

    • The presence of aneuploidy in IC bladder cells could signify an underlying epithelial cell abnormality.
    • Further research is warranted to elucidate the role of aneuploidy in interstitial cystitis pathogenesis.
