Summary

Head-down tilt in rats reduces heart polypeptide initiation. This process is mediated by heat-shock cognate/heat-shock protein-70 (HSC/HSP-70) facilitating eukaryotic initiation factor-2 alpha (eIF-2 alpha) phosphorylation.

Background:

  • Previous studies showed head-down tilt decreases heart polypeptide initiation rate in rats.

Purpose of the Study:

  • To investigate the role of HSC/HSP-70 and eIF-2 alpha phosphorylation in mediating the decrease in heart polypeptide initiation during head-down tilt.
  • To quantify changes in eIF-2 alpha phosphorylation and HSC/HSP-70 levels under head-down tilt conditions.

  • Two-dimensional gel electrophoresis was used to analyze eIF-2 alpha phosphorylation.
  • Polysomal HSC/HSP-70 levels and polysome center-of-mass were assessed at different time points during head-down tilt.
