Summary

Astrocyte cells in different brain regions show varied responses to noradrenaline (NA) and glutamate (GLU). Cerebellar astrocytes exhibit the strongest cyclic GMP (cGMP) response to NA, indicating a key role for alpha 1-adrenoceptors and nitric oxide synthase in this region.

Background:

  • Previous studies demonstrated nitric oxide (NO)-dependent cyclic GMP (cGMP) formation in response to noradrenaline (NA) and glutamate (GLU) in rat cerebrum astrocyte cultures.
  • Astrocyte heterogeneity in agonist responses across different brain regions was previously suggested but not fully characterized.

Purpose of the Study:

  • To investigate the heterogeneity of astrocyte responses to NA and GLU in cultures from cerebellum, hippocampus, and cortex.
  • To elucidate the specific signaling pathways involved in NA-stimulated cGMP formation in cerebellar astrocytes.

Main Methods:

  • Primary astrocyte-enriched cultures were established from rat cerebellum, hippocampus, and cortex.
  • cGMP levels were measured following stimulation with NA, GLU, and the NO donor sodium nitroprusside (SNP).

  • Pharmacological agents including L-arginine analogues, prazosin, and the calcium ionophore A23187 were used to probe signaling pathways. Calcium dependency was assessed by omitting calcium from the medium.
  • Main Results:

    • Significant heterogeneity in agonist responses was observed: NA response was highest in cerebellar, intermediate in hippocampal, and lowest in cortical astrocytes. GLU had minimal effects in cortical and cerebellar cultures.
    • SNP produced higher cGMP levels than agonists, with efficacy order cerebellum > cortex > hippocampus. Responses to NA and SNP in cerebellar astrocytes were sensitive to culture conditions.
    • NA-induced cGMP formation in cerebellar astrocytes was blocked by L-arginine analogues and prazosin, mimicked by A23187, and dependent on extracellular calcium, suggesting alpha 1-adrenoceptor and calcium-dependent NO synthase involvement.

    Conclusions:

    • Astrocyte responses to neurotransmitters like NA and GLU are region-specific.
    • Cerebellar astrocytes possess functional alpha 1-adrenoceptors and calcium-dependent nitric oxide synthase (cNOS) mediating NA-stimulated cGMP production.
    • These findings support an astroglial localization of alpha 1-adrenoceptors and cNOS involved in NA signaling pathways in the cerebellum.
