Summary

Chronic nonsteroidal anti-inflammatory drug (NSAID) use in patients with gallstones was associated with reduced gallbladder mucin. This suggests NSAIDs may play a role in preventing gallstone formation in humans.

Background:

  • Gallbladder mucin secretion is key in gallstone formation.

Purpose of the Study:

  • To investigate the association between chronic NSAID use and gallbladder bile mucin content and lipid profiles in humans.
  • To determine if NSAID use affects gallstone pathogenesis in human gallbladder bile.

Main Methods:

  • Retrospective analysis of 230 morbidly obese patients undergoing gastric bypass surgery.
  • Categorization into four groups: gallstones with NSAID use, gallstones without NSAID use, no gallstones with NSAID use, and no gallstones without NSAID use.
  • Direct aspiration of gallbladder bile for mucin and lipid analysis.

  • Patients with gallstones had significantly higher gallbladder mucin concentrations than those without gallstones (0.897 +/- 0.226 vs 0.173 +/- 0.039 mg/ml).
  • Among patients with gallstones, chronic NSAID users exhibited reduced gallbladder mucin levels compared to non-users (1.18 +/- 0.43 vs 0.74 +/- 0.19 mg/ml).

Conclusions:

  • Chronic NSAID use is linked to lower gallbladder mucin concentrations in patients with gallstones.
  • These findings support a potential role for NSAIDs in mitigating gallstone formation in humans by reducing mucin content.
