    Background:

    • The maternal immune system must tolerate the semi-allogeneic fetus.
    • The placenta plays a critical role in mediating maternal-fetal tolerance.

    Purpose of the Study:

    • To investigate the placenta's role as an immunoabsorbent for maternal antibodies targeting fetal antigens.
    • To determine if placental absorption of antibodies is dependent on fetal circulation.

    Main Methods:

    • In vivo administration of 125I-labeled antibodies against murine major histocompatibility complex (MHC) to pregnant females.
    • Quantification of antibody uptake in placental tissue and maternal organs at different gestational ages (13 and 17 days).
    • Assessment of placental antibody absorption in surgically decerebrated 13-day pregnant mice.

    Main Results:

    • The placenta selectively absorbed anti-MHC antibodies when the fetus expressed the target antigen.
    • Differential placental uptake was observed as early as 2 hours and as late as 12 hours post-injection.
    • Antibody absorption occurred in placentas even after surgical removal of the fetus, indicating fetal circulation is not required.
    • Specificity controls ruled out Fc receptor binding or hybrid vigor effects.
    • No significant increase in antibody binding was observed in maternal livers, spleens, kidneys, or lungs.

    Conclusions:

    • The placenta functions as a paternal antigen-bearing immunoabsorbent.
    • This placental immunoabsorbent capacity may act as a barrier, protecting the semi-allogeneic fetus from maternal immune attack.
    • The findings highlight a novel mechanism for immune tolerance during pregnancy.
