Summary

Glutamine supplementation in diets significantly reduces methotrexate clearance and renal elimination in rats. This dietary change increases overall methotrexate exposure for both host and tumor tissues.

Area of Science:

  • Pharmacology
  • Gastroenterology
  • Nutritional Science

Background:

  • Methotrexate is a crucial chemotherapy agent with known gastrointestinal toxicities.
  • Glutamine-supplemented diets have shown promise in mitigating methotrexate-induced enterocolitis.
  • The impact of glutamine on methotrexate's pharmacokinetic profile remains incompletely understood.

Purpose of the Study:

  • To investigate the effect of glutamine supplementation on the pharmacokinetics of methotrexate.
  • To determine how glutamine influences the formation of 7-hydroxy-methotrexate, a key metabolite.
  • To assess changes in methotrexate's systemic clearance and renal elimination under glutamine-rich diets.

Main Methods:

  • Adult female Lewis rats were divided into glycine-supplemented (control) and glutamine-supplemented diet groups.

Related Experiment Videos

  • Animals received a 10 mg/kg dose of methotrexate.
  • Serum methotrexate pharmacokinetics and renal elimination were measured in separate subgroups.
  • Main Results:

    • Glutamine supplementation led to a 25% decrease in mean methotrexate total serum clearance.
    • Renal elimination of methotrexate was significantly reduced by 65% in the glutamine group.
    • No significant alterations were observed in methotrexate's volume of distribution or half-life.

    Conclusions:

    • Dietary glutamine significantly decreases systemic clearance and renal elimination of methotrexate.
    • This reduction in clearance results in increased methotrexate exposure for both the host and tumor.
    • Further research is warranted to explore the clinical implications of glutamine supplementation on methotrexate therapy.
