Abstract:
The radioreceptor method is sensitive, simple, and specific, requiring little, or no purification of sample prior to analysis. It is possible to process hundreds of samples in a matter of hours (sample throughput may be enhanced through the use of a cell harvester for rapid filtration of bound [3H]cAMP). Assays for endogenous cAMP and cAMP measured using a radioactive precursor give similar results in terms of fold stimulation. The main advantage of the former technique lies in the use of cells or tissues with poor uptake of [3H]adenine.
