Summary

Methotrexate (MTX) showed beneficial effects for most patients with ankylosing spondylitis (AS) who did not respond to other treatments. Side effects were mild and reversible, indicating MTX is a viable treatment option.

Treatment of active rheumatoid arthritis with leflunomide: two year follow up of a double blind, placebo controlled trial versus sulfasalazine.

Annals of the rheumatic diseases·2001

Area of Science:

  • Rheumatology
  • Pharmacology

Background:

  • Ankylosing spondylitis (AS) is a chronic inflammatory disease.
  • Treatment options for AS include NSAIDs and sulfasalazine, but some patients do not respond adequately.

Purpose of the Study:

  • To evaluate the efficacy and toxicity of methotrexate (MTX) in patients with active ankylosing spondylitis.
  • To assess MTX as a treatment for patients refractory to NSAIDs and sulfasalazine.

Main Methods:

  • An open-label, single-observer study involving 11 patients with active AS.
  • Patients received oral methotrexate (7.5-15 mg weekly) for at least 24 weeks.
  • Efficacy was measured by changes in assessed variables and patient evaluation.

Main Results:

Related Experiment Videos

  • Significant improvement in assessed variables was observed.
  • Four patients continued MTX, with some reducing NSAID dosage.
  • Mild and reversible side effects were reported; 3 patients discontinued MTX due to disease flares but later restarted.
  • Conclusions:

    • Methotrexate demonstrated beneficial effects for the majority of ankylosing spondylitis patients studied.
    • MTX appears to be an effective and well-tolerated treatment option for AS patients unresponsive to conventional therapies.
