Summary

Beta-amyloid precursor protein (beta APP) is present in healthy human digital skin nerves and cells, but beta-amyloid peptide (beta/A4) is not detected. This study maps beta APP distribution in glabrous skin.

Background:

  • Beta-amyloid precursor protein (beta APP) and beta-amyloid peptide (beta/A4) are implicated in neurological disorders.
  • Their presence and function in healthy human skin are not well understood.

Purpose of the Study:

  • To investigate the occurrence and distribution of beta APP and beta/A4 in healthy adult human digital skin.
  • To determine the cellular localization of these proteins within the skin.

Main Methods:

  • Immunoblotting and immunohistochemical techniques were employed.
  • Analysis was performed on whole skin, dermis, and epidermis samples.
  • Antibodies specific to beta APP and beta/A4 were used to detect protein presence and localization.

  • Beta APP-like proteic bands were detected in skin and dermal extracts, including full-length isoforms.
  • Beta APP immunoreactivity was observed in dermal nerves, Meissner's and Pacinian corpuscles, blood vessels, sweat glands, and epidermis.
  • Beta/A4 was not detected by immunoblotting, and its immunoreactivity mirrored beta APP but was not extracellular.

Conclusions:

  • Beta APP is normally present in human glabrous (digital) skin, particularly within neural and cellular structures.
  • Beta/A4 is not detected in healthy digital skin under normal conditions.
  • These findings suggest a potential physiological role for beta APP in skin.
