Insights
Researchers characterized the O-polysaccharide from enteroaggregative Escherichia coli strain 73-1, a cause of childhood diarrhea. The study determined its structure is composed of repeating pentasaccharide units.
Area of Science:
- Microbiology
- Carbohydrate Chemistry
- Structural Biology
Background:
- Enteroaggregative Escherichia coli (EAEC) is a significant cause of bacterial diarrhea, particularly in children.
- Lipopolysaccharide (LPS) is a major component of the outer membrane of Gram-negative bacteria, including E. coli.
- The O-polysaccharide (O-antigen) is a key structural and antigenic component of LPS.
Purpose of the Study:
- To elucidate the detailed chemical structure of the O-polysaccharide from a specific Chilean EAEC strain (73-1) isolated from a diarrheal patient.
- To contribute to the understanding of the structural diversity of E. coli O-antigens.
Main Methods:
- Isolation and purification of the lipopolysaccharide (LPS) from E. coli strain 73-1.
- Chemical analyses including sugar composition and methylation analysis of the polysaccharide.
- Advanced spectroscopic techniques, specifically 1H-NMR and 13C-NMR spectroscopy, were employed.
Main Results:
- The O-polysaccharide was found to be constructed from repeating pentasaccharide units.
- Detailed structural information regarding the specific arrangement of sugars and linkages within the repeating unit was obtained through spectroscopic data.
Conclusions:
- The study successfully determined the repeating unit structure of the O-polysaccharide from E. coli strain 73-1.
- This structural characterization provides valuable data for understanding the antigenic properties and potential pathogenicity of this EAEC strain.
Abstract:
The polysaccharide part of the lipopolysaccharide obtained from an enteroaggregative Escherichia coli strain isolated from a young child with diarrhoea in Santiago, Chile (strain 73-1) was investigated. Sugar and methylation analyses of native and partially degraded polysaccharide together with 1H-NMR and 13C-NMR spectroscopy revealed that the polysaccharide is built of pentasaccharide repeating units. The structure of the repeating unit of E. coli strain 73-1 O-polysaccharide is (formula: see text)
