[Effect of nitrates on the dynamics of methemoglobin formation in rats of various ages]
Abstract:
Wistar male rats with body mass of 50-83 g and 327-365 g were treated per os with solution of sodium nitrate in single dose of 500 or 1000 mg/kg of body mass (on the base of nitrate-ion). The level of methemoglobin (MH) and activity of methemoglobinreductase (MHR) in blood were analyzed. It was shown that level of MH in blood of young immature rats was increased with appearance of two peaks after ingestion both doses of nitrate. The appearance of two peaks of MH in mature rats was observed only after the high dose of nitrate. Low dose of nitrate caused increase of MH concentration only one time. Both doses of nitrate did not effect on MHR activity in rat blood.
Insights
Sodium nitrate ingestion increased methemoglobin (MH) in young rats at both doses. Mature rats showed a dual MH increase only with high doses, with no effect on methemoglobinreductase (MHR) activity.
Area of Science:
- Toxicology
- Biochemistry
- Pharmacology
Context:
- Sodium nitrate exposure can occur through diet and environmental sources.
- Methemoglobinemia is a condition where methemoglobin levels increase in the blood.
- Understanding nitrate toxicity is crucial for public health and risk assessment.
Purpose:
- To investigate the dose-dependent effects of sodium nitrate on methemoglobin levels in Wistar rats of different age groups.
- To assess the impact of sodium nitrate on methemoglobinreductase activity.
- To compare the toxicological response to nitrate in immature versus mature rats.
Summary:
- Administration of sodium nitrate (500 or 1000 mg/kg) to Wistar male rats resulted in increased methemoglobin (MH) levels.
- Young, immature rats exhibited a biphasic increase in MH after both low and high nitrate doses.
- Mature rats showed a biphasic MH increase only with the high dose, while the low dose caused a single MH elevation. Methemoglobinreductase (MHR) activity remained unaffected in both age groups and across all doses.
Impact:
- This study highlights age-dependent differences in susceptibility to nitrate-induced methemoglobinemia.
- Findings contribute to understanding the toxicokinetics and toxicodynamics of nitrate exposure.
- Provides data for establishing safe exposure limits and informing clinical management of nitrate poisoning.
