Summary

Cyclic AMP-reactive proteins (cARP) were found in whole saliva and parotid fluid, with higher concentrations in parotid fluid. Auditory stimuli increased cARP levels in saliva, while phosphodiesterase (PDE) activity was detected only in whole saliva.

Cyclic AMP-receptor proteins in human salivary glands.

Background:

  • Regulatory (R) subunits of cyclic AMP-dependent protein kinase (PKA) are known to be present in parotid fluid.
  • Secretory cyclic nucleotide phosphodiesterase (PDE) activity in saliva has not been previously characterized.

Purpose of the Study:

  • To compare cyclic AMP-reactive proteins (cARP) and PDE activity in human whole saliva versus parotid fluid.
  • To investigate if environmental stimuli alter secretory cARP levels.

Main Methods:

  • Non-invasive collection of whole and parotid ductal saliva from healthy volunteers.
  • Photoaffinity labeling with [32P]-8-N3-cAMP followed by autoradiography to identify R subunits.
  • Measurement of cyclic nucleotide PDE activity via conversion to tritiated 5'-nucleotide.

Main Results:

  • Type II cyclic AMP-kinase (cAPK) R subunits (RII, RIIa) were detected in all parotid saliva samples.
  • Whole saliva contained RII in over 95% of samples, but at 50-90% lower concentrations than parotid fluid.
  • Auditory stimuli (60-80 dB) caused a 2- to 5-fold increase in cARP in both sexes (p < 0.05).
  • PDE activity was measurable in whole saliva but not detected in parotid fluid.
