Summary

Sumatriptan, a 5-HT receptor agonist, altered neuroendocrine function in healthy males. It reduced prolactin and increased growth hormone, but did not affect cortisol levels.

Purpose of the Study:

  • To investigate the neuroendocrine effects of sumatriptan in healthy male subjects.
  • To compare the effects of sumatriptan with placebo.

Main Methods:

  • A placebo-controlled, cross-over study design was employed.
  • 11 healthy male subjects received sumatriptan (6 mg subcutaneously) or placebo.
  • Plasma levels of prolactin, growth hormone, and cortisol were measured.

Main Results:

  • Sumatriptan significantly decreased plasma prolactin levels compared to placebo.
  • Sumatriptan significantly increased plasma growth hormone levels compared to placebo.
  • No significant effect of sumatriptan on plasma cortisol concentrations was observed.

Conclusions:

  • Sumatriptan exerts distinct neuroendocrine effects, differing from other 5-HT receptor agonists.
  • These effects may be due to selective activation of 5-HT1D or 5-HT1B receptors.
  • Nonspecific stress responses or alterations in pituitary blood flow cannot be ruled out as contributing factors.
