Purpose of the Study:

  • To investigate the chemosensitivity of nociceptive afferents to L-glutamate.
  • To determine if L-glutamate contributes to inflammation-related pain.

Main Methods:

  • Used an isolated spinal-cord-tail preparation of neonatal rats.
  • Applied L-glutamate and other chemicals to rat tail skin.
  • Measured nociceptive reflexes.

Main Results:

  • L-glutamate stimulated nociceptive reflexes in a dose-dependent manner (ED50 = 136 microM).
  • Other L-amino acids and D-glutamate did not stimulate reflexes.
  • Known algogens like bradykinin and acetylcholine also stimulated reflexes, while histamine and substance P did not.

Conclusions:

  • L-glutamate selectively activates nociceptive afferents.
  • L-glutamate released from macrophages may contribute to inflammation-induced pain by stimulating cutaneous nociceptors.
