Summary

MHC class II molecule costimulation enhances T cell growth and activation. This effect is observed through interleukin-2 and T cell receptor/CD3 pathways, interacting with CD28 signals.

Area of Science:

  • Immunology
  • Cell Biology

Background:

  • MHC class II-positive T cells are implicated in autoimmune disorders.
  • Class II molecule stimulation activates protein tyrosine kinases and modulates T cell responses.

Purpose of the Study:

  • To investigate the role of MHC class II molecules in regulating T cell growth.
  • To determine how class II costimulation affects T cell activation pathways.

Main Methods:

  • Utilized immobilized HLA-DR monoclonal antibody (mAb) for costimulation.
  • Assessed interleukin (IL)-2-supported T cell growth and T cell receptor/CD3-induced activation.
  • Examined the interaction with CD28 accessory signals and protein kinase C activation.

Main Results:

  • Costimulation of class II molecules significantly enhanced IL-2-supported T cell growth in most CD4+, CD45RAlow, ROhigh T cell lines.

Related Experiment Videos

  • Class II costimulation also enhanced TCR/CD3-induced T cell activation and potentiated CD28-mAb-induced sensitivity to protein kinase C.
  • Responsiveness to class II costimulation was independent of T cell cytokine production profiles (TH1-like or THO-like).
  • Conclusions:

    • MHC class II costimulation is a potent enhancer of T cell activation via TCR/CD3 and IL-2 pathways.
    • Class II costimulation interacts with CD28 signals to promote the growth of allospecific T cell lines.
    • These findings highlight a significant regulatory role for MHC class II molecules in T cell proliferation and activation.
