Summary

Krox-20 is essential for peripheral nervous system myelination. Disruption of Krox-20 in mice prevents Schwann cells from completing myelination, blocking this crucial developmental process.

Induction of Myelin Gene Expression in Murine Schwann Cells in Primary Culture and in a Schwann Cell Line.

Annals of the New York Academy of Sciences·2017

Area of Science:

  • Neuroscience
  • Molecular Biology
  • Developmental Biology

Background:

  • Myelination by Schwann cells is vital for peripheral nerve function.
  • The precise molecular controls governing myelination remain incompletely understood.
  • Krox-20, a transcription factor, is known to influence embryonic development.

Purpose of the Study:

  • To investigate the role of the Krox-20 gene in Schwann cell differentiation and myelination.
  • To determine if Krox-20 is essential for the completion of peripheral nervous system myelination.

Main Methods:

  • Generation of a Krox-20 null allele in mice using gene targeting with the lacZ gene.
  • Analysis of Krox-20 expression in Schwann cells during development.
  • Phenotypic characterization of Krox-20 deficient (Krox-20-/-) mice, including myelination status and gene expression.

Related Experiment Videos

Main Results:

  • Krox-20 is expressed in Schwann cells prior to myelination.
  • Krox-20 deficiency leads to an arrest in Schwann cell differentiation.
  • Krox-20 null mice exhibit incomplete myelination, with reduced wrapping and absence of late myelin genes (e.g., protein zero, myelin basic protein).

Conclusions:

  • Krox-20 is a critical regulator of Schwann cell differentiation and myelination.
  • Krox-20 controls downstream genes necessary for the maturation of myelin in the peripheral nervous system.
  • Understanding Krox-20's function provides insight into the molecular basis of myelination disorders.
