Area of Science:

  • Histopathology
  • Cell Biology
  • Biochemistry

Context:

  • Archival tissues often present challenges for accurate protein visualization.
  • Standard silver staining methods for nucleolar organizer regions proteins (AgNORs) can yield variable results.
  • Formalin fixation and paraffin embedding can obscure cellular substructures.

Purpose:

  • To improve the visualization of AgNORs in formalin-fixed and paraffin-embedded archival tissues.
  • To establish a reproducible and artifact-free silver staining protocol for AgNORs.
  • To facilitate standardized AgNOR quantification on historical tissue samples.

Summary:

  • A novel wet autoclave pretreatment method was applied to formalin-fixed and paraffin-embedded archival tissues.
  • This pretreatment substantially improved the visualization of AgNORs, yielding results comparable to alcohol-fixed tissues.

Related Experiment Videos
