Summary

Human Leukocyte Antigen-G (HLA-G) exhibits more genetic diversity than previously thought. This Human Leukocyte Antigen gene variation suggests HLA-G may play a broader role in immune responses during pregnancy.

Background:

Human Leukocyte Antigen-G (HLA-G) is a non-classical MHC class I molecule with restricted tissue expression.
Its unique localization at the maternal-fetal interface suggests a crucial role in successful human pregnancy.
Previous research indicated a limited allelic repertoire for HLA-G compared to classical HLA genes.

Purpose of the Study:

To comprehensively assess the sequence variation within the first six exons of the HLA-G gene.
To investigate the potential for HLA-G to present diverse peptides and elicit allogeneic responses.

Main Methods:

Analysis of nucleotide variation in the first six exons of the HLA-G gene.
Examination of samples from 45 healthy African American individuals.
Utilized sensitive techniques for detecting nucleotide substitutions (>95% detection rate).

Main Results:

Identified two sequence variations in exon 2 (alpha-1 domain) leading to amino acid substitutions.
Detected 24 sequence variations in exon 3 (alpha-2 domain) resulting in amino acid substitutions.
These findings reveal significant sequence diversity in HLA-G.

Conclusions:

HLA-G possesses a greater degree of polymorphism than previously recognized.
The observed sequence variations indicate HLA-G's potential to present a wide array of peptides.
This diversity suggests HLA-G may elicit allogeneic responses, similar to classical HLA class I genes, impacting pregnancy and transplantation immunology.
