Summary

Human heat shock proteins (HSPs) are differently distributed in ocular cicatricial pemphigoid (OCP) compared to healthy or atopic conjunctiva. OCP shows increased HSP expression in the stroma and vasculature, suggesting a role in inflammation.

Background:

  • Ocular cicatricial pemphigoid (OCP) is a chronic inflammatory disease affecting the conjunctiva.
  • Heat shock proteins (HSPs) are involved in cellular stress responses and inflammation.

Purpose of the Study:

  • To investigate the distribution of specific human heat shock proteins (HSPs) including HSP90, HSP70 (inducible and constitutive forms), HSP65, and HSP27.
  • To compare HSP expression in conjunctival biopsy specimens from patients with OCP, atopic keratoconjunctivitis, and healthy individuals.

Main Methods:

  • Conjunctival biopsy specimens were obtained from ten OCP patients, ten atopic keratoconjunctivitis patients, and ten healthy individuals undergoing cataract surgery.
  • An immunoperoxidase technique was employed using monoclonal antibodies against human HSPs to analyze protein distribution.

  • Normal and atopic keratoconjunctivitis epithelia showed significant expression of HSP90 and HSP27, with lesser amounts of HSP70 and HSP65.

  • Upregulated expression of HSP90, constitutive HSP70, and HSP27 in the OCP stroma suggests a role in inflammation, potentially driven by cytokine release.
  • The prominent presence of HSPs in the conjunctival vascular endothelium of OCP patients indicates a previously unrecognized involvement of the vasculature in the disease pathogenesis.
