Summary

The X Chromosome controlling element locus (Xce) influences X inactivation in mice. New microsatellite markers near the Xist gene show strong linkage with Xce, but with allelic differences suggesting they may not be the same genetic element.

Background:

  • The X Chromosome controlling element locus (Xce) is a key regulator of X inactivation in mice.
  • The Xist gene is located in the central region of the X Chromosome and is also implicated in X inactivation.

Purpose of the Study:

  • To investigate the relationship between the Xce locus and the Xist gene.
  • To isolate and characterize microsatellite markers near the Xist locus to assess their linkage with Xce.

Main Methods:

  • Isolation of three microsatellite markers from a YAC containing the Xist locus.
  • Analysis of linkage and linkage disequilibrium between these markers and Xce in various mouse strains.
  • Typing of the Xceb strain JU/Ct using the flanking microsatellite markers.
