Summary

Lamotrigine effectively reduced seizures and improved well-being in girls with Rett syndrome by inhibiting glutamate release. Further controlled studies are warranted for this neurological disorder.

Background:

  • Rett syndrome is a rare genetic neurodevelopmental disorder.
  • Elevated cerebrospinal fluid (CSF) glutamate levels are implicated in Rett syndrome pathophysiology.
  • Glutamate excitotoxicity is a potential mechanism contributing to neurological dysfunction.

Observation:

  • Lamotrigine, an anticonvulsant, is known to inhibit glutamate release.
  • A small cohort of four girls diagnosed with Rett syndrome received Lamotrigine as an add-on therapy.

Findings:

  • All four patients experienced a seizure reduction of 50% or more.
  • Patients also demonstrated an improved overall well-being following Lamotrigine treatment.
  • The results suggest a positive therapeutic response to Lamotrigine in Rett syndrome.

Implications:

  • Lamotrigine shows promise as a treatment for seizure management in Rett syndrome.
  • Inhibition of glutamate release may be a viable therapeutic strategy.
  • Early-stage controlled studies are recommended to validate these preliminary findings.
