Summary

Adoptive immunotherapy using autologous activated lymphocytes (AAL) shows promise for treating resistant cancers. Early addition of Interleukin-2 (IL-2) to lymphocyte cultures significantly enhances their tumor-killing ability.

Background:

  • Adoptive immunotherapy offers a treatment option for malignant tumors resistant to conventional therapies.
  • Tumor-infiltrating lymphocytes (TIL) are effective but often unavailable; autologous activated lymphocytes (AAL) serve as an alternative.
  • Current AAL protocols involve leukapheresis, activation, and support with IL-2 and cimetidine.

Purpose of the Study:

  • To optimize the yield and activation of AAL for improved efficacy.
  • To investigate the impact of timing of Interleukin-2 (IL-2) addition on lymphocyte activation and function.
  • To enhance the tumor-specificity of AAL through in vitro priming techniques.

  • AAL were generated and cultured with varying schedules of IL-2 addition.
  • Expression of activation markers (CD25, HLA-DR) and tumor cell killing capacity were assessed.
