Summary

Normal human corneas contain activated T-lymphocytes. These CD8+ cytotoxic T-cells, identified via flow cytometry, may explain the eye's unique immune properties.

Background:

  • The human cornea's immune privilege is well-established.
  • However, the specific T-lymphocyte populations and their activation states within the normal cornea remain incompletely understood.

Purpose of the Study:

  • To characterize the phenotype and function of T-lymphocytes present in normal human corneal tissue.
  • To investigate whether these corneal T-lymphocytes are activated and possess cytotoxic potential.

Main Methods:

  • Human corneal tissues from 10 donors were cultured in Interleukin-2 (IL-2) medium.
  • Flow cytometry (FACS) analysis was performed using monoclonal antibodies against various T-cell markers (CD3, CD4, CD8, TCR alpha/beta, TCR alpha/delta) and activation markers (HLA-DR, CD25).
  • Functional T-cell proliferation assays (PLT) were conducted.

Main Results:

  • T-cell growth was more frequent from peripheral corneal regions compared to central regions.
  • The majority of cultured T-cells expressed the CD8 marker, indicating cytotoxic T-lymphocytes.
  • These CD8+ cells were activated (HLA-DR+) and demonstrated cytotoxic function (CD18+).
  • All T-cell receptors were of the alpha/beta type, and no Natural Killer (NK) cells were detected.

Conclusions:

  • Normal human corneas harbor activated, cytotoxic CD8+ T-lymphocytes.
  • These findings suggest a role for these lymphocytes in the immunological distinctiveness of the ocular environment.
