Summary

Baseline lipid peroxide levels are not elevated in hypercholesterolemic individuals compared to controls. This suggests that increased susceptibility to atherosclerosis in hypercholesterolemia is not due to higher baseline lipid peroxide levels.

Purpose of the Study:

  • To determine if baseline lipid peroxide levels are higher in hypercholesterolemic subjects compared to normolipidemic controls.
  • To rigorously control for in vitro oxidation during sample processing.

Main Methods:

  • Two assay methods were used: thiobarbituric acid (TBA) and hemoglobin-methylene blue (HbMB).
  • Plasma and serum were collected and processed under strict conditions to prevent in vitro oxidation.
  • Lipid peroxide determination was performed within 2 hours of blood collection.

  • Lipid peroxide levels were significantly higher in serum than in plasma for both hypercholesterolemic and control groups.
  • No significant difference in lipid peroxide levels was observed between hypercholesterolemic and control subjects in either plasma or serum.

Conclusions:

  • The increased tendency towards atherosclerosis in hypercholesterolemia is unlikely related to baseline plasma lipid peroxide levels.

  • Further research is needed to elucidate the mechanisms of atherosclerosis in hypercholesterolemia.
