Background:

  • Depression treatment has evolved, with newer antidepressant classes emerging.
  • Tricyclic antidepressants (TCAs) were historically dominant, but newer agents offer potentially improved profiles.

Purpose of the Study:

  • To compare the efficacy and safety of newer antidepressants against traditional TCAs in acute depression therapy.
  • To evaluate mianserin, moclobemide, and selective serotonin reuptuptake inhibitors (SSRIs) versus TCAs.

Main Methods:

  • A meta-analysis was conducted on studies of acute depression therapy (first 8 weeks).
  • Clinical efficacy was measured using the Hamilton Rating Scale for Depression.
  • Comparisons included TCAs, mianserin, moclobemide, and specific SSRIs (citalopram, fluoxetine, fluvoxamine, paroxetine, sertraline).
